Background

MAOB (Monoamine Oxidase B) is one of two neighboring mitochondrial enzyme family members which catalyze the oxidative deamination of amines, such as dopamine, norepinephrine, and serotonin. The other member is MAOA. MAOB preferentially degrades benzylamine and phenylethylamine. MAOB catalyzes the oxidative deamination of biogenic and xenobiotic amines and plays an important role in the metabolism of neuroactive and vasoactive amines in the central nervous system and peripheral tissues.

Application Notes

Recombinant MAOB protein is suitable for use in the study of enzyme kinetics, inhibitor screening, and selectivity profiling.

Monoamine Oxidase Assay: 40 uM substrate was incubated with MAOB protein in reaction buffer for 1 hour at room temperature, then the product was incubated with Fluorescence for 0.5 hour at temperature. The activity of MAOB was detected by Promega MAO-GloTM Assay.

Protein Details

Recombinant MAOB (accession number NP_000889.3) was expressed in Sf9 cells and contains an N-terminal FLAG-Tag with an observed molecular weight of 59.8 kDa. The recombinant protein is >85% pure by SDS-PAGE.

Recombinant MAOB protein gel.
MAOB protein was run on an 8% SDS-PAGE gel and stained with Coomassie blue.

Recombinant MAOB activity assay.
40 uM substrate was incubated with MAOB protein in reaction buffer for 1 hour at room temperature, then the product was incubated with Fluorescence for 0.5 hour at temperature. The activity of MAOB was detected by Promega MAO-GloTM Assay.

Storage

Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
