Immunogen

This IκBα phospho Ser32,36 antibody was raised against a synthetic peptide containing phosphorylated serines at amino acid residues 32 and 36 of human IκBα.

Buffer

PBS containing 0.02% sodium azide. Sodium azide is highly toxic.

 

IκBα phospho Ser32,36 mAb tested by Western blot.
Jurkat cells were treated for 30 minutes with 100 µg/ml ALLN (N-Acetyl-Leu-Leu-Norleucinal), a calpain and proteasome inhibitor that prevents IκBα dephosphorylation, followed by incubation with (lanes 2 & 4) or without 1 nM TNF-α (1 & 3). The membranes were blotted with the anti-phospho-IκBα (lanes 1 & 2) or anti-IκBα, which recognizes both phosphorylated and non-phosphorylated forms of IκBα. The data shows that IκBα phospho Ser32,36 mAb detects specifically the phosphorylated forms of IκBα.

Background

IκBα phospho Ser32,36NFκB (NFκB p50 & NFκB p65) signaling is controlled to a large extent by the sequestration of the NFκB complex in the cytoplasm by its association with one of the IκB family of proteins. IκBα is phosphorylated at Ser32 and Ser36 by the IκB Kinase (IKK) complex, resulting in the degradation of IκB and the nuclear translocation of NFκB.

Positive Control

Extracts from Jurkat cells treated for 30 minutes with 100 µg/ml ALLN (N-Acetyl-Leu-Leu-Norleucinal), followed by incubation with or without 1 nM TNF-α cell extract can be used as a positive control for IκBα phospho Ser32, 36 antibody.
