Applications

ChIP Validated ChIP-Seq Validated Western Blot Validated

Application Notes

Applications Validated by Active Motif:
ChIP: 10 µl per ChIP
ChIP-Seq:10 µl each
WB*: 1:500 - 1:1,000 dilution

ChIP-Seq validation was performed by Active Motif's Epigenetics Services. The complete data set is available in the UCSC Genome Browser by clicking here.

*Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western blot.

Published Applications

The following applications have been published using this antibody. Unless noted above, Active Motif may not have validated the antibody for use in these applications:

Immunogen

This BRD8 / SMAP2 antibody was raised against a peptide corresponding to amino acids within the N-terminal region of human BRD8.

Buffer

Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

BRD8 / SMAP2 antibody (pAb) tested by ChIP-Seq.
ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 30 ug of chromatin from an adenocarcinoma cell line and 10 µl of antibody. ChIP DNA was sequenced on the Illumina HiSeq and 20 million sequence tags were mapped to identify BRD8 / SMAP2 binding sites. The image shows binding across a region of chromosome 9. You can view the complete data set in the UCSC Genome Browser, starting at this specific location, here.

Background

BRD8 (Bromodomain-containing protein 8, SMAP2, TRCP120) is a member of the Bromodomain-containing family of chromatin associated proteins. BRD8 is a member of the NuA4 histone acetyltransferase complex, involved in the transcriptional activation of specific genes through the acetylation of histones H2A and H4. BRD8 contains two Bromodomains, a motif that has been demonstrated to facilitate interaction with acetylated histones.

Positive Control

Active Motif's HeLa nuclear extract (Catalog No. 36010) can be used as a positive control for BRD8 / SMAP2 antibody.

Chromatin IP Assays

This antibody has been validated for use in ChIP and/or ChIP-Seq, and can be used with Active Motif's ChIP-IT® High Sensitivity Kit or our magnetic bead-based ChIP-IT® Express Kits.
