Background

BRDT is similar to the RING3 protein family. It possesses two bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain functions as a ‘reader’ of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets. It shows binding specificity for acetylated H2BK12/15, H3K9/14/18/23, H4K12, H4K5/8, H4K8/12, H4K12/16, H4K5/12, H4K5/8/12/16 and H4K5/8/12/16.

Recombinant BRDT (21-137) protein, GST-Tag that includes amino acids 21-137 (Bromodomain) of human BRDT protein (accession number NP_001229735.2) was expressed in E.coli and contains an N-terminal GST Tag with a molecular weight of 40.2 kDa.

HTRF assay for recombinant BRDT (21-137) protein, GST-Tag activity
1 µM H4K5/8/12/16(ac4) peptide was incubated with different concentrations of BRDT (21-137) protein, GST-tag in a 10 µl reaction system containing 50 mM HEPES-NaOH pH 7.4, 0.1% BSA for 1 hour, then 10 µl GST antibody and SA-XL665 mixture (each 1:100 dilution in the reaction buffer) was added to each reaction system and incubated for 30 min. All the operations and reactions were performed at room temperature. HTRF assay was used for detection.
